Kit contents

The kit includes all reagents required for DNA cleanup:
1. Binding buffer - na osnove chaotropic agents i auxiliary components dlya dissolution agarose gel i sorbtsii NA na filtre kolonki;
2. Wash buffer - dlya promyvki nucleic acids ot ostatkov agarose gel i komponentov reaktsionnykh smesey;
3. Elution buffer - dlya elyutsii i khraneniya NA, pH=8,9;
4. Spin-kolonki - dlya filtratsii reaktsionnoy smesi i sorbtsii NA.

Instructions for use

DNA cleanup procedure

Sample preparation (gel)
1. Vyrezat uchastok gelya s nuzhnym fragmentom DNA i place ego v predvaritelno vzveshennuyu tube, dalee vzvesit tube s proboy i vyschitat raznitsu. Ves gelya v mg priravnivaetsya k ego obemu v uL (1 mg gelya = 1 uL gelya).
2. V tube add 3 obema svyazyvayushchego bufera (ot obema gelya), no ne menee 300 uL.
3. Incubate smes v termostate pri 55-58℃ until complete dissolution gelya, periodicheski peremeshivaya on a vortex mixer.
Optsionalno: dlya fragmentov DNA menee 500 bp posle dissolution gelya vnesti v reaktsiyu 1 obem izopropanola (ot obema gelya) i peremeshat. Dalee pereyti k etapu sorbtsii i promyvki.
Sample preparation (reaktsionnaya smes)
1. Esli obem RS sostavlyaet menee 25 uL, to neobkhodimo dovesti ego do 50 uL vodoy ili TE-buferom i prinyat dannyy obem za osnovnoy. K obemu RS add 3 obema svyazyvayushchego bufera, peremeshat.
Optsionalno: dlya fragmentov DNA menee 500 bp vnesti v reaktsiyu 2 obema izopropanola (ot obema RS). Dalee pereyti k etapu sorbtsii i promyvki.
Sorbtsiya i promyvka NA
1. Peremestit ves obem smesi v spin-kolonku.
2. Centrifuge v techenie 30 sekund pri 10-13 tys. ob/min, udalit filtrat iz sobiratelnoy tubes.
3. Add v spin-kolonku 750 uL promyvochnogo bufera.
4. Centrifuge v techenie 30 sekund pri 10-13 tys. ob/min, udalit filtrat iz sobiratelnoy tubes.
5. Centrifuge spin-kolonku v techenie minuty pri 10-13 tys. ob/min.
6. Peremestit spin-kolonku v novuyu tube 1,5-2 mL. Sobiratelnuyu tube utilizirovat.
Nucleic acid elution
1. Add na tsentr membrany spin-kolonki 50-100 uL elyuiruyushchego bufera.
2. Incubate v techenie 3-10 minut at room temperature.
3. Centrifuge v techenie 30-60 sekund pri 10-13 tys. ob/min.
4. Utilizirovat spin-kolonki.
Optsionalno: dlya povysheniya vykhoda NA filtrat mozhno povtorno nanesti na membranu kolonki i/ili ispolzovat nagretyy do 60-70℃ elyuiruyushchiy bufer.
Poluchennye rastvory nucleic acids can be stored do 7 dney pri temperature ot +2℃ do +4℃ i do dvukh let pri temperature -20℃. Po absorbance ratio na A260/A280 purity of the resulting solution DNA corresponds to ≥1,7.

Storage conditions

Buffers nabora "Amplex" can be stored pri temperature ot +4℃ do +25℃ for 12 months from the manufacturer's release date. If a precipitate is present pri minimalnykh temperaturakh khraneniya, warm the solutions to room temperature.

  7. Spin-kolonki s sobiratelnymi probirkami (prisutstvuyut v komplekte);
  8. Vorteks;
  9. Dry block thermostat s vozmozhnostyu podderzhaniya temperaturnogo rezhima v diapazone 25-80ºS dlya probirok obemom 1,5-2 mL;
  10. High-speed microcentrifuge dlya probirok obemom 1,5-2 mL do 13 tys. ob/min.
